高效液相色谱法测定互叶白千层中没食子酸的含量

摘要:
为了建立用高效液相色谱测定互叶白千层中没食子酸含量的方法,为互叶白千层的质量标准制定提供方法和依据,采用高效液相色谱法(HPLC)测定互叶白千层中没食子酸含量。色谱柱为Diamonsil C18(4.6mm×250mm,5μm),流动相为乙腈-0.2%磷酸(3:97),检测波长274nm,柱温为室温,流速1ml/min。结果,没食子酸在0.2016~1.2096μg范围内呈良好线性关系(r=0.9991),平均回收率为101.87%,RSD为1.76%(n=9)。本方法简便可行,重现性好,可以作为互叶白千层中没食子酸含量的测定方法。

Abstract:
A high-performance liquid chromatography(HPLC) method was developed for the determination of gallic acid in Melaleuca alternifolia, which provided the basis for establishing the quality standard of Melaleuca alternifolia.The determination was performed on Diamonsil C18(4.6mm×250mm, 5μm) column with acetonitrile-0.2% phosphoric acid(3:97) as mobile phase with flow rate of 1.0 ml/min at detection wavelength of 274 nm.And the column temperature was set at room temperature.The average recovery rate(n=9) of gallic acid was 101.87% and RSD was 1.76%.This method is simple, rapid, accurate and reliable, and is suitable for the quality control of gallic acid in Melaleuca alternifolia.
