福建新记录种全缘燕尾蕨的鉴定及其叶绿体基因组基本特征

摘要:
本研究报道了全缘燕尾蕨(Cheiropleuria integrifolia)在福建木兰溪源省级自然保护区的分布,充实了福建省蕨类植物区系中双扇蕨科(Dipteridaceae) 的种类。通过野外调查,提供了该种野外照片及局部特征,并对其生境进行了简要分析,凭证标本保存于福建师范大学植物标本馆(Herbarium of Fujian Normal University,FNU)。另外,使用Illumina NovaSeq 6000平台对该物种进行完整叶绿体基因组测序,后进行组装、注释,并基于rbcL基因序列构建系统进化树。系统进化树分析表明燕尾蕨属(Cheiropleuria)与双扇蕨属(Dipteris)聚为一支(BP/PP:100/1.00),全缘燕尾蕨与燕尾蕨(C.bicuspis)聚为一独立小支(BP/PP:100/1.00),且均为强支持。全缘燕尾蕨叶绿体基因组为典型的四分体结构,总长度为153 682 bp,包含124个基因(31个tRNA基因、8个rRNA基因和85个蛋白质编码基因)。结合形态特征和分子鉴定结果,可判定该物种为全缘燕尾蕨。研究结果增加了双扇蕨科的叶绿体基因组数据,为后续研究提供科学依据。

Identification and Chloroplast Genome Characterization of a Newly Recorded Species Cheiropleuria integrifolia in Fujian Province

Abstract:
The distribution of Cheiropleuria integrifolia in the Fujian Provincial Mulanxi Source Nature Reserve was reported,enriching the species of Dipteridaceae in the fern flora of Fujian Province.Field surveys were conducted,providing field photos of this species,along with the descriptions of local features and brief introduction of the habitats.The voucher specimen was preserved in the Herbarium of Fujian Normal University.In addition,Illumina NovaSeq 6000 was used to sequence the whole chloroplast genome of this species,followed by assembly,annotation,and construction of a phylogenetic tree based on rbcL.The phylogenetic tree revealed that Cheiropleuria and Dipteris clustered into one clade (BP/PP:100/1.00),while C.integrifolia and C.bicuspis formed a smaller clade (BP/PP:100/1.00),both of which were strongly supported.The chloroplast genome of C.integrifolia was a typical tetrad structure with a total length of 153 682 bp and comprising 124 genes (31 tRNA genes,8 rRNA genes,and 85 protein-coding genes).Based on both the morphological characteristics and molecular evidence,this species was identified as C.integrifolia.These findings not only contribute valuable chloroplast genome data to the study of Dipteridaceae but also establish a scientific foundation for future investigations in this field.
