海洋热带假丝酵母产角鲨烯发酵工艺研究

摘要:
[目的]为了从海洋热带假丝酵母(Candida tropicalis)菌株中获取高产量的角鲨烯活性物质。[方法]研究不同碳源、氮源、培养温度、培养基初始pH值、培养时间、光照和氧气等条件对海洋热带假丝酵母菌株产角鲨烯的影响。[结果]确定最优培养基为(W/V)1%酵母膏+2%蛋白胨+2%蔗糖,最佳摇瓶发酵条件:有氧条件下,初始pH值为5.0,培养温度25℃,经48 h培养,角鲨烯产量可达2.25 mg/g干重。[结论]采用优化后工艺条件进行发酵,获得的角鲨烯产量明显提高。

Study on Fermentation Process for Squalene Production by Marine Candida tropicalis

Abstract:
[Objective] Optimization experiment was performed in order to obtain high yield of squalene active substances from marine Candida tropicalis strain.[Methods] This study compared the influence of different carbon source, nitrogen source, culture temperature, initial pH, incubation time, the illumination and oxygen condition on the yield of squalene from marine Candida tropicalis strain.[Results] The optimal medium (W/V) was 1% yeast extract, 2% peptone and 2% sucrose.The best conditions for fermentation in shaking flask were the optimal initial pH at 5.0 and the optimal temperature at 25℃.Under aerobic conditions, the squalene content from the strain reached up to 2.25 mg/g(dry weight) after 48 hours of culture.[Conclusion] This strain can produce higher content of squalene active substance on the conditions obtained from the optimal fermentation process.
