小月柳珊瑚中生物碱类化学成分及其抗骨肉瘤细胞增殖活性研究

摘要:
[目的]研究小月柳珊瑚(Menella kanisa)中生物碱类化学成分及其抗骨肉瘤细胞增殖活性。[方法]采用多种分离方法和波谱分析方法,对小月柳珊瑚中生物碱类化学成分进行分离和结构鉴定。采用MTT法测试部分生物碱类化合物抗骨肉瘤细胞U2OS增殖活性。[结果]从小月柳珊瑚中分离出7个化合物,分别鉴定为6-(1'-purine-6',8'-dionyl) suberosanone (1)、1-(3'-carbonylbutyl) purine-6,8-dione (2)、腺嘌呤脱氧核苷(3)、次黄嘌呤(4)、7-甲基腺嘌呤(5)、胸腺嘧啶脱氧核苷(6)、尿嘧啶脱氧核苷(7)。化合物12对骨肉瘤细胞U2OS增殖显示出抑制活性,IC50值分别为13.3 μg/mL和55.0 μg/mL。[结论]化合物1~7均首次从小月柳珊瑚中分离得到,其中化合物12的抗骨肉瘤细胞活性也为首次报道。

Abstract:
[Objective] The alkaloids from marine gorgonian Menella kanisa and their activities of anti-proliferation on U2OS cancer cell were investigated.[Methods] The alkaloids were isolated and purified by column chromatography on silica gel, and their structures were identified by spectral analyses and compared with literatures.The cytotoxicity of compounds 1~2 toward human osteosarcoma cell U2OS cancer cell line was evaluated by MTT methods.[Results] Seven alkaloids were obtained from M.kanisa and characterized as 6-(1'-purine-6', 8'-dionyl)suberosanone (1), 1-(3'-carbonylbutyl)purine-6, 8-dione (2), adenine deoxyribonucleoside (3), hypoxanthine (4), 7-methyladenine (5), thymine deoxyribonucleoside (6) and uracil deoxyriboside (7).Compounds 1 and 2 showed moderate cytotoxicity against osteosarcoma U2OS cell line with IC50 of 13.3 and 55.0 μg/mL, respectively.[Conclusion] Compounds 1~7 were isolated from gorgonian M.kanisa for the first time.The antitumor activity of Compounds 1 and 2 against U2OS in vitro was also reported for the first time.
