应用PCR-SSCP和免疫组化法检测29例广西南部肝癌(HCC)组织中的N-ras基因突变和HBV感染状况。结果,肝癌中N-ras基因在第2~37密码子之间的突变率为79.3%,其中22例(75.86%)有2~5个突变位点。该基因突变也见于癌旁组织(80.77%)。肝组织中HBsAg和HBxAg检出率分别为86.2%和79.3%,两者具有相关性,并与N-ras基因突变率呈相平行的趋势。因广西南部的肝癌与AFB1污染有关,本研究中HCC的N-ras基因的突变可能也与AFB1的作用有关。 |

Abstract: |
The mutation of N-ras gene and hepatitis B virus (HBV) markers were detected in 29 cases of hepatocellular carcinomas (HCC) in Southern Guangxi by polymerase chain reaction-single strand conformation polymorphism (PCR SSCP) and immunohistochemistry.The aberrations at N-ras codons 2~37 were found at 79.31% in HCC. 2~5 mutated codons of N-ras gene were tested in 22 cases (75.86%) HCC.These mutations were also found in 80.77% liver tissue surrounding the HCCs.The HBsAg and HBxAg positive rates in liver tissue were 86.2% and 79.3% respectively.There was a positive correlation between the detection of HBsAg and HBxAg,on the other hand,there was a parallel tendency between the HBV markers and mutation rate of N-ras gene.Since the aflatoxin B1 (AFB1) contamination is a risk factor for HCC in Southern Guangxi,AFB1 may play a role the mutation of N-ras gene. |
