超声辅助低共熔溶剂提取当归药渣粗多糖的工艺优化

摘要:
研究当归药渣提取粗多糖工艺,以当归药渣粗多糖(ARP)的提取率为指标,考察低共熔溶剂的种类、摩尔比、水含量、提取温度、提取时间和液料比对ARP提取率的影响,并结合响应面分析法优化提取工艺参数,得到最优提取工艺。结果表明:氯化胆碱和乙二醇组成的低共熔溶剂更适合提取ARP。最优提取工艺为:提取温度82℃,提取时间40 min,摩尔比6,ARP提取率为14.43%±0.1%,ARP总糖含量为47.54%±1.23%。相比传统的热水提取ARP提取率为9.85%,其ARP总糖含量为25.45%±1.37%。因此,低共熔溶剂是一种较好的提取溶剂,可替代传统溶剂进行多糖的提取,具有实际应用价值。

Optimization of Deep Eutectic Solvent-based Ultrasound-assisted Extraction of Crude Polysaccharides from Angelica Residue

Abstract:
The extraction rate of Angelica Residue Crude Polysaccharide (ARP) was used as the index to investigate the effects of different the type of deep eutectic solvents (DES), mole ratio, water content, extraction temperature and extraction time, ratio of water to materials on yield of ARP. The extraction process parameters were optimized by response surface methodology, and the optimal extraction process was obtained. The results showed that DES composed of choline chloride and ethylene glycol was proved to be suitable for the extraction. The optimum extraction process as follows: extraction temperature of 82 ℃, the extraction time of 44.74 min, and the mole ratio of 6. Under the optimal extraction conditions, yield of ARP was 14.43%±0.1%, and the total sugar content was 47.54%±1.23%.Compared with hot water extraction, yield of ARP was 9.85%, and the total sugar content was 25.45%±1.37%. Therefore, DES were an excellent extraction solvent alternative to the extraction of polysaccharides, and has a practical value.
