福建新记录种黎平景天形态学鉴定及其叶绿体基因组特征

摘要:
为探究福建分布新记录种黎平景天(Sedum lipingense)的分类学地位。本文对黎平景天的形态学特征进行描述,并结合核ITS序列系统发育分析与叶绿体基因组数据,对其分类鉴定与基因组特征进行研究。形态学鉴定结果支持福建采集的材料为黎平景天;基于ITS序列构建的系统发育树显示:本研究获得的序列与NCBI数据库中黎平景天序列聚类高度一致,且获得强支持(BP/PP=99/1.00),进一步证实两者为同一物种。利用Illumina NovaSeq 6000平台对该物种叶绿体基因组进行测序,组装与注释后的结果表明:黎平景天基因组为典型环状四分体结构,全长149 919 bp,含130个基因(85个蛋白质编码基因、8个rRNA基因和37个tRNA基因)。上述结果丰富了福建省景天属植物多样性,同时也可为基于叶绿体基因组数据的景天属系统演化研究提供参考。

Abstract:
To clarify the taxonomic status of Sedum lipingense, a newly recorded species in Fujian Province, we documented its morphological characters and conducted integrative taxonomic analyses combining nuclear ITS-based phylogenetics with chloroplast genome data. Morphological evidence supports that the Fujian collections are referable to S. lipingense. The phylogenetic tree inferred from ITS sequences showed that the sequences generated in this study clustered tightly with published S. lipingense sequences deposited in NCBI, with strong support (BP/PP = 99/1.00), further confirming their conspecificity. The complete chloroplast genome was sequenced using the Illumina NovaSeq 6000 platform and subsequently assembled and annotated. The plastome exhibits a typical circular quadripartite structure and is 149,919 bp in length, comprising 130 genes (85 protein-coding genes, 8 rRNA genes, and 37 tRNA genes). These results expand our knowledge of Sedum diversity in Fujian and provide genomic resources for future studies on phylogeny and evolutionary history of Sedum based on chloroplast genome data.
