本研究旨在探讨遗传因素及其与环境因素的交互作用对广西红水河流域老年人群认知功能的影响。研究纳入895名广西红水河流域的老年人,采用病例-对照设计,将其分为认知正常组和认知损害组进行比较。通过提取外周血DNA检测味觉相关的单核苷酸多态性(SNP)位点,评估各SNP位点与味觉感知、认知状态的关联,并采用广义多因子降维法(GMDR)分析SNP-SNP及SNP-环境交互效应对认知状态的影响。结果显示,多个SNP位点与老年人味觉功能显著相关,苦味受体基因TAS2R38 rs10246939位点的突变等位基因C在显性遗传模型中是认知功能的风险因素,T/C和C/C基因型个体的患病风险高于T/T型;而CLEC5A rs1285950位点C/A基因型相较于A/A基因型在认知损害组中的比例更高,C/A基因型可能是认知损害的风险因素。GMDR分析显示,AGK、DENND11和CLEC5A基因的四基因座模型以及包含味觉功能、性别的SNP-环境交互模型均对认知功能具有良好的预测效果。综上,广西红水河流域老年人群认知功能受多基因遗传因素影响,味觉相关SNP位点及其与味觉功能、性别等环境因素的交互作用在其中发挥重要作用。 |

The purpose of this study was to explore the effects of genetic factors and their interaction with environmental factors on the cognitive function of the elderly in the Hongshui River Basin. A total of 895 elderly individuals from the Hongshui River Basin were included. A case-control design was used to divide them into cognitive normal group and cognitive impairment group for comparison.The Single Nucleotide Polymorphism (SNP) loci related to taste perception were detected by extracting peripheral blood DNA,and the association of each SNP locus with taste perception and cognitive status was evaluated.The Generalized Multi-factor Dimensionality Reduction method (GMDR) was used to analyze the effects of SNP-SNP and SNP-environment interaction on cognitive status.The results showed that multiple SNP loci were significantly correlated with the taste function of the elderly.The mutant allele C of the bitter taste receptor gene TAS2R38 rs10246939 locus was a risk factor for cognitive function in the dominant genetic model,individuals with the T/C and C/C genotypes have a higher risk of disease than ones with the T/T genotype,while the C/A genotype of CLEC5A rs1285950 locus was higher in the cognitive impairment group than the A/A genotype.The C/A genotype might be a risk factor for cognitive impairment. GMDR analysis showed that the four-locus model of AGK,DENND11 and CLEC5A genes and the SNP-environment interaction model including taste function and gender all had good predictive effects on cognitive function.In summary,the cognitive function of the elderly populations in the Hongshui River Basin is affected by multi-gene genetic factors,and taste-related SNP loci and their interactions with environmental factors such as taste function and gender play a significant role in it. |
