野生玫瑰(Rosa rugosa Thunb.)具有重要经济、生态和育种价值,被列为国家二级重点保护野生植物。迁地保护对野生植物遗传多样性恢复有重要作用,而开展迁地保护评估可为后续保护工作提供科学指导。本研究基于SSR分子标记对野生玫瑰开展迁地保护成效评估,结果显示:野生种群中以辽宁大连种群的遗传多样性最高,而吉林珲春种群最低。野生种群的总遗传变异Ht为0.3834、种群内遗传变异Hs为0.2803、种群间遗传分化系数Fst为0.2689,遗传变异主要存在于种群内。各种群观测杂合度均小于期望杂合度,杂合子缺失(Fis > 0),群体间亲缘关系较近,存在一定程度的近交。合并后的迁地保护种群的等位基因数(Na)、观测杂合度(Ho)、期望杂合度(He) 、Shannon 信息指数(I)等均高于野生种群。以上结果说明,迁地保存种群遗传多样性高于野生种群,野生玫瑰的迁地保护对其种群遗传多样性恢复有正向作用,迁地保护成效较好,可为其他野生植物开展迁地保护提供借鉴。 |

Wild rose (Rosa rugosa Thunb.) has important economic, ecological and breeding values, and is listed as a national second-class key protected wild plant. Ex situ conservation plays an important role in the restoration of wild plant genetic diversity, and ex situ conservation assessment can provide scientific guidance for subsequent conservation work. In this study, the effects of ex situ conservation of wild rose were evaluated based on SSR molecular markers. The results showed that the genetic diversity of the wild rose population in Dalian, Liaoning province was the highest, while that in Hunchun, Jilin Province was the lowest. The total genetic variation (Ht) of wild population was 0.3834, the in-population genetic variation (Hs) was 0.2803, and the coefficient of inter-population genetic differentiation (Fst) was 0.2689. The observed heterozygosity was lower than the expected heterozygosity, heterozygote loss (Fis>0), close relationship between populations, and some degree of inbreeding. The number of alleles (Na), observed heterozygosity (Ho), expected heterozygosity (He) and Shannon information index (I) of the combined ex situ protected population were higher than those of the wild population. These results indicate that the genetic diversity of ex situ preserved population is higher than that of wild population, ex situ conservation of wild rose has a positive effect on the restoration of genetic diversity of its population, and ex situ conservation has a good effect, which can provide a reference for other wild plants to carry out ex situ conservation. |
