茉莉酸甲酯对条斑紫菜贝壳丝状体壳孢子放散的影响

摘要:
茉莉酸甲酯(MJ)是普遍存在于植物体内的生长调节物质,在促进植物的生长发育和成熟衰老等方面发挥重要作用。为了探讨MJ对条斑紫菜贝壳丝状体放散期生理和分子机制方面的作用,本文比较了不同浓度的MJ对条斑紫菜贝壳丝状体放散量与叶绿素荧光参数Fv/Fm。研究发现,1-50 μmol/L MJ能够促进贝壳丝状体壳孢子放散并且导致放散高峰提前,其中10 μmol/L MJ效果最显著。1-9 d,10 μmol/L MJ组的Fv/Fm普遍低于对照组,呈现出先降低后升高的趋势。利用转录组学方法,分析了MJ对壳孢子放散期条斑紫菜贝壳丝状体多种代谢途径的影响,10 μmol/L MJ组相较对照组共筛选出482个差异性表达基因,其中上调175个,下调307个,KEGG差异性表达基因最显著富集的途径分别为糖酵解/糖异生、碳固定等。荧光定量PCR (qRT-PCR) 验证结果显示,5个与糖酵解、碳固定相关的差异性表达基因的转录表达上调,与高通量测序结果一致,表明MJ通过能量利用和碳固定促进了壳孢子放散。

Effect of methyl jasmonate on release process of the conchocelis of Neopyropia yezoensis

Abstract:
Methyl jasmonate (MJ) is an ubiquitous growth regulator in plants, plays an important role in growth and development of plant, as well as ripening and senescence . In order to explore the physiological and molecular mechanisms of MJ on the release of conchocelis from Neopyropia yezoensis, the effects of different concentrations of MJ on the release amount of conchocelis and Fv/Fm were compared. It was found that 1-50 μmol/L MJ could promote the release of conchospores and lead to the early release peak, of which 10 μmol/L MJ had the most significant effect. From 1 to 9 d, the Fv/Fm value in the 10 μmol/L MJ group was generally lower than that of the control group, showing a trend of first decreasing and then increasing. The effects of MJ on multiple metabolic pathways of conchocelis during the release period of N. yezoensis were studied by transcriptomic methods. A total of 482 differentially expressed genes were screened in the 10 μmol/L MJ group compared with the control group, of which 175 were up-regulated and 307 were down-regulated. KEGG significantly enriched metabolic pathways such as glycolysis/gluconeogenesis, and carbon fixation, with 4 genes under the condition of 10 μmol/L MJ, the transcription level was significantly upregulated, indicating that MJ promoted the release of conchospores through energy utilization and carbon fixation.
