瑶药黄红钻对CIA模型大鼠关节炎的治疗作用研究

摘要:
目的 探究黄红钻水提物对胶原诱导关节炎(Collagen-induced Arthritis,CIA)模型大鼠的治疗作用。方法 采用牛Ⅱ型胶原诱导建立CIA大鼠模型,口服灌胃给药,观察黄红钻水提物高、中、低剂量组(10.8、5.4、2.7 g/kg)对CIA大鼠体重、足部肿胀程度、关节炎指数评分、踝关节病理形态及血清肿瘤坏死因子-α(Tumor necrosis factor-α,TNF-α),白细胞介素6(Interleukin-6,IL-6),白介素-1β(Interleukin-1β,IL-1β),IL-1β表达水平的影响。结果 与空白组比较,模型组大鼠在初次免疫第8天体重显著升高(P<0.05),实验结束时体重无显著性差异,足部肿胀程度以及关节炎指数评分显著升高(P<0.01),踝关节软骨层和软骨下层骨组织受到重度侵蚀,血清炎症因子IL-1β、IL-6及TNF-α水平显著升高(P<0.01);与模型组相比,黄红钻高剂量组大鼠在初次免疫第8天体重显著升高(P<0.05),实验结束时黄红钻低、中、高剂量组大鼠体重均无显著差异,阳性药组及黄红钻各剂量组大鼠的右后爪心肿胀度显著下降(P<0.05),但黄红钻中剂量组在第29天右后爪心肿胀度下降速度减慢,阳性药组及黄红钻各剂量组大鼠的关节炎指数出现下降趋势,其中在第22天阳性药组和黄红钻低、高剂量组大鼠显著下降(P<0.05);阳性药组,黄红钻水提物低、中、高剂量组的踝关节炎症细胞浸润及纤维组织增生情况均有减轻,其血清炎症因子IL-1β、IL-6及TNF-α的水平显著下降(P<0.05 P<0.01)。结论 黄红钻水提物可以显著降低CIA大鼠血清炎症因子IL-1β、IL-6及TNF-α的水平,从而缓解CIA大鼠体内的炎症反应。

Study on the Therapeutic Effect of Jasminum pentaneurum Hand.-Mazz on Rheumatoid Arthritis in CIA Rats

Abstract:
Objective To study the therapeutic effect of water extract of Jasminum pentaneurum Hand.-Mazz on collagen-induced arthritis (CIA) model rats. Methods The CIA rat model was induced by bovine type II collagen, and the effects of water extract of J.pentaneurum in high-dose, medium-dose and low-dose groups (10.8, 5.4, 2.7 g/kg) on body weight, foot swelling, arthritis index score, ankle pathological morphology and expression levels of serum TNF-α, IL-6, and IL-1β in CIA rats were observed, respectively. Results Compared with the blank group, the weight of rats in the model group increased significantly on the day 8 of primary immunization (p<0.05), but there was no significant difference in weight at the end of the experiment, the degree of foot swelling and the score of arthritis index increased significantly (p<0.01), the cartilage layer and subcartilage bone tissue of the ankle joint were severely eroded and the level of serum inflammatory factor IL-1β,IL-6 and TNF-α increased significantly (p<0.01). Compared with the model group, the weight of rats in the high dose group of J.pentaneurum increased significantly on the day 8 of the initial immunization (p<0.05), and there was no significant difference in the weight of rats in the low, medium and high dose groups of J.pentaneurum. The swelling degree of the right posterior paw heart of rats in the positive drug group and each dose group of Jasminum pentaneurum decreased significantly (p<0.05), but the decrease rate of swelling degree of the right posterior paw heart of rats in the medium dose group of J.pentaneurum slowed down on the day 29, The arthritis index of rats in the positive drug group and the J.pentaneurum dose groups showed a downward trend, in which the positive drug group and the J.pentaneurum low and high dose groups significantly decreased on the day 22 (p<0.05). In the positive drug group, the inflammatory cell infiltration and fibrous tissue hyperplasia of ankle joint were reduced in the low, medium and high dose groups of J.pentaneurum water extract, and the levels of serum inflammatory cytokines IL-1β, IL-6 and TNF-α were significantly decreased (P<0.05 and P<0.01). Conclusion The water extract of J.pentaneurum could significantly reduce the level of serum inflammatory factors including IL-1β, IL-6 and TNF-α in CIA rats, and thus the related inflammatory reaction was alleviated.
