基于指纹图谱-网络药理学-多指标定量的黄槿 抗炎质量标志物研究

摘要:
为筛选黄槿的质量标志物(quality marker,Q-Marker)并建立其含量测定方法,本研究采用高效液相色谱法建立22批黄槿的指纹图谱,运用多种化学模式识别技术对数据进行分析;并构建“疾病-成分-靶点-通路”网络模型预测Q-Marker,进而对其含量进行测定。结果显示本研究建立了22批黄槿指纹图谱,共标定12个共有峰,经分析筛选3个差异成分,预测芦丁、异槲皮苷、山柰酚-3-O-芸香糖苷为黄槿Q-Marker候选成分。网络药理学分析显示,芦丁、异槲皮苷、山柰酚-3-O-芸香糖苷可能通过表皮生长因子受体、丝裂原活化蛋白激酶3与肿瘤坏死因子等核心靶点,调控白细胞介素-17、EGFR酪氨酸激酶耐药性抑制剂及松弛素等信号通路,从而发挥抗炎作用。该质量评价方法准确可行,基于指纹图谱和网络药理学筛选的质量标志物,可为完善黄槿质量标准提供依据,并为其抗炎药效物质基础及作用机制提供理论支撑。

Research on Quality Marker for Inflammation of Hibiscus tiliaceus Based on Fingerprint,Network Pharmacology and Multi-index Quantitative Analysis

Abstract:This study aims to screen the quality markers(Q-Marker) of Hibiscus tiliaceus and establish its quantitative analysis methods.The fingerprints of 22 batches of Hibiscus tiliaceus were established by high-performance liquid chromatography and subsequently analyzed using chemical pattern recognition technologies.The network diagram of “disease-components-targets-pathways” was constructed to predict the Q-Marker and quantitative analysis was conducted simultaneously. The results showed that the fingerprints of 22 batches of Hibiscus tiliaceus were established,and total of 12 common peaks were calibrated.Three differential components were obtained after analysis.It was predicted that rutin,isoquercitrin and kaempferol-3-O-rutinoside were candidate components of quality markers of Hibiscus tiliaceus .Network pharmacology analysis revealed that rutin,isoquercitrin and kaempferol-3-O-rutinoside might exert therapeutic effects on inflammation through core targets,including epidermal growth factor receptor,mitogen-activated protein kinase 3 and tumor necrosis factor,influencing signaling pathways such as interleukin-17,EGFR tyrosine kinase inhibitor resistance and relaxin.The established quality evaluation method demonstrates high accuracy and reliability.The Q-Markers screened based on fingerprint and network pharmacology can provide a basis for improving the quality control of Hibiscus tiliaceus, as well as offer theoretical support for its anti-inflammatory pharmacologic material basis and mechanism of action.
