| 为揭示越南卷柏Selaginella pseudotamarscina叶绿体基因组特征及其系统位置,本研究基于高通量测序数据,从头组装了越南卷柏完整的叶绿体基因组,并对其基因组结构、基因构成及基本信息进行分析;结合其他卷柏科Selaginellaceae物种的叶绿体基因组数据,进行基因组比较分析并构建系统发生树。研究结果表明越南卷柏叶绿体基因组为正向重复(DR)结构,全长125 931 bp,总GC含量为53.3%,RNA编辑位点预测数量高达1 545个,共编码82个基因,包括65个蛋白编码基因、9个转运RNA(tRNA)基因和8个核糖体RNA(rRNA)基因。叶绿体基因组比较分析结果表明,越南卷柏的微卫星(简单重复序列,SSR)数量和类型最少,仅有两个二核苷酸重复序列;其和近缘种之间无结构重排现象,重复区边界变化较大;核苷酸变异主要集中在单拷贝区的基因间隔区,共鉴定出5个高变区域;系统发生树强烈支持越南卷柏与S.imbricata聚为一支,不属于旱生卷柏S.stauntoniana分支。本研究丰富了莲座状复苏卷柏的遗传资源信息,并为理解卷柏科植物叶绿体基因组的进化提供了重要理论参考。 |

| To uncover the chloroplast genome characteristics and taxonomic position of Selaginella pseudotamarscina,this study assembled a complete chloroplast genome based on high-throughput sequencing data.The genome structure,gene composition,and basic information of S.pseudotamarscina were analyzed.The genomic comparative analysis with other close relative species was performed and a phylogenetic tree was built.The results indicated that the chloroplast genome of S.pseudotamarscina had the Direct Repeat (DR) structure,a total length of 125 931 bp,GC content of 53.3%,and 1 545 predicted RNA editing sites.It encoded 82 genes,including 65 protein-coding genes,9 transfer RNA (tRNA) genes,and 8 ribosomal RNA (rRNA) genes.Comparative analysis of chloroplast genomes showed that S.pseudotamarscina had the fewest microsatellite (Simple Sequence Repeat,SSR),with only two dinucleotide repeat sequences.There was no structural rearrangement between S.pseudotamarscina and its close relatives,while there were significant variations in the boundaries of repeat regions.The nucleotide variation regions were mainly concentrated in the gene spacer region of single-copy regions,and five highly variable regions were identified.The phylogenetic tree strongly supported the clade formed by S.pseudotamarscina with S.imbricata,indicating that it did not belong to the S.stauntoniana clade.This study enriches the genetic resource information of the rosette and resurrection species of Selaginella and provides theoretical references for understanding the evolution of chloroplast genomes in Selaginella plants. |
