白花泡桐对Zn的耐受性及耐性株系筛选研究

摘要:
为了筛选白花泡桐耐Zn优良株系,本研究利用植物组织培养技术,将白花泡桐组培苗分别接入含Zn 2.0、4.0、6.0、8.0、10.0 mmol/L五个浓度梯度的生根培养基中培养60 d,观测白花泡桐对Zn的耐受情况并筛选半致死浓度;在此基础上,将白花泡桐74个不同株系组培苗接种至含半致死浓度Zn的生根培养中培养60 d后,观测记录了各株系的根、茎、叶生长或死亡参数,并以这些生长参数作为耐Zn性评价依据,采用隶属函数分析法对不同株系进行耐Zn性综合评价。结果表明,白花泡桐耐Zn性较强,在Zn 2.0 mmol/L时其根、茎、叶的生长几乎未受影响;白花泡桐组培小苗对Zn的半致死浓度为5.34 mmol/L,在此浓度下,从74个不同的白花泡桐株系中筛选到6个耐Zn株系。上述结果为重金属污染土壤修复提供一定的物质基础。

Study on Tolerance of Paulownia Fortunei to Zn and Screening of Resistant Strains

Abstract:
In order to screen excellent Zn-tolerant lines of Paulownia Fortunei, tissue culture seedlings of Paulownia Fortunei were cultured in root medium with five concentration gradients of Zn 2.0, 4.0, 6.0, 8.0 and 10.0 mmol/L, respectively, for 60 days by using plant tissue culture technology and the tolerance of Paulownia Fortunei to Zn was observed and the semi-lethal concentration was screened. After the semi-lethal concentration was determined, 74 different tissue culture seedlings were inoculated into root culture containing semi-lethal concentration of Zn for 60 days, and the growth or death parameters of roots, stems and leaves of each line were observed and recorded. Based on these growth parameters, the Zn tolerance of different strains was evaluated by membership function analysis. The results showed that Paulownia Fortunei had strong Zn tolerance, and the growth of roots, stems and leaves was almost unaffected when Zn was 2.0 mmol /L. The semi-lethal concentration of Zn for Paulownia Fortunei was 5.34 mmol/L, and 6 Zn-tolerant lines were screened from 74 different Paulownia Fortunei lines. These results provide a material basis for the remediation of heavy metal contaminated soil.
