玛卡调控5-HT、DA对小鼠自闭症样行为及神经环路的影响

摘要:
通过研究玛卡调控5-羟色胺(5-hydroxytryptamine,5-HT)、多巴胺(Dopamine,DA)对小鼠自闭症样行为及相关神经环路的影响,初步探讨玛卡治疗自闭症的疗效及其作用机制。在整个实验中将ICR小鼠及C57/BL6小鼠分别随机设空白对照组及玛卡低、中、高剂量组,采用灌胃给药的方式,并通过高架十字迷宫实验、旷场实验及三腔社交实验,观察玛卡给药后两种不同种系小鼠行为学的变化;采用酶联免疫试剂盒检测小鼠血浆5-HT及DA含量;通过c-fos蛋白检测相关脑区的神经元活性。结果显示,与空白对照组比较,玛卡提高了ICR小鼠在高架十字迷宫实验中进入开放臂的时间、次数及指数(P<0.05或P<0.01),减少了其在旷场实验中的总移动距离(P<0.05或P<0.01),提高了其在三腔社交实验中的社会性指数、社会新颖性指数与社会记忆指数(P<0.05),并提高了其血浆5-HT及DA含量(P<0.05或P<0.01或P<0.001);c-fos检测结果显示,VTA、VP、NAcc、mPOA及amygdala阳性细胞数目明显增多(P<0.05或P<0.01或P<0.001)。本研究结果提示,玛卡对ICR小鼠自闭症样行为有明显的改善作用,且该作用可能是通过调节5-HT及DA的水平并影响相关的神经环路实现的。

Effects of Maca by Regulating 5-HT and DA on Autism-like Behavior and Neural Circuit in Mice

Abstract:
By studying the effects of Maca regulating 5-hydroxytryptamine (5-HT) and dopamine (DA) on autism-like behavior and related neural circuits in mice,the efficacy and mechanism of Maca in the treatment of autism were preliminarily discussed.In the whole experiment,ICR mice and C57/BL6 mice were randomly divided into blank control group and Maca low-,medium- and high-dose groups.The drug was administered by intragastric gavage,and the elevated cross labyrinth test,open field test and three-chamber social test were carried out to observe the behavioral changes of two different strains of mice after Maca administration.The contents of 5-HT and DA in plasma of mice were detected by enzyme-linked immunosorbent assay kit.The activity of neurons in related brain regions was detected by c-fos protein.The results showed compare with the blank control group,Maca increased the time,times and index of entering the open arm in the elevated cross labyrinth test of ICR mice (P<0.05 or P<0.01),decreased the total distance in the open field test (P<0.05 or P<0.01),and increased the social index,social novelty index and social memory index in the three chambers test (P<0.05).The contents of 5-HT and DA in plasma were increased (P<0.05 or P<0.01 or P<0.001).The results of c-fos detection showed that the number of positive cells in VTA,VP,NAcc,mPOA and amygdala increased significantly (P<0.05 or P<0.01 or P<0.001).The results of this study suggest that Maca has a significant improvement effect on autism-like behavior in ICR mice,and this effect may be achieved by regulating the levels of 5-HT and DA and affecting the related neural circuits.
