厚藤提取物对胶原诱导关节炎模型大鼠的治疗作用研究 |

摘要: |
为探究厚藤(Ipomoea pes-caprae)提取物对胶原诱导关节炎(Collagen-induced Arthritis,CIA)模型大鼠的治疗作用,本研究采用牛Ⅱ型胶原诱导建立CIA大鼠模型,观察厚藤提取物高、中、低剂量组(2.271 2,1.135 6,0.567 8 g/kg)对CIA大鼠踝关节肿胀、足跖肿胀、关节炎症评分、踝关节病理切片及血清中前列腺素E2(Prostaglandin E2,PGE2)、类风湿因子(Rheumatoid Factor,RF)、C-反应蛋白(C-reactive Protein,CRP)表达水平的影响。结果表明:与模型组相比,给药干预后,踝关节肿胀度、足跖肿胀度有减轻趋势但无显著性差异(P>0.05);厚藤提取物高、中剂量组能显著降低关节炎症指数、PGE2、CRP水平(P<0.05,P<0.01);厚藤提取物各剂量组均能显著降低RF水平(P<0.01)。踝关节病理切片结果显示,厚藤提取物高、中剂量组病变程度相比于模型组均得到减轻。综上可知,厚藤提取物对CIA大鼠具有较好的治疗作用,能减轻关节肿胀、缓解滑膜炎症状,这可能与减少PGE2、RF、CRP表达有关。 |

In order to explore the therapeutic effect of Ipomoea pes-caprae extracts (IPE) on collagen-induced arthritis (CIA) model rats,CIA rat model was established by bovine type Ⅱ collagen in this study.The effects of high,medium and low doses of I.pes-caprae extracts (2.271 2,1.135 6,0.567 8 g/kg) on the ankle swelling,footpad swelling,joint inflammation score,pathological section of ankle joint and serum prostaglandin E2 (PGE2),rheumatoid factor (RF),C-reactive protein (CRP) expression levels of CIA rats were observed.The results showed that compared with the model group,the ankle swelling degree and plantar swelling degree showed a decreasing trend but no significant difference after administration intervention (P>0.05).Moreover,the high-dose and medium-dose IPE groups could significantly reduce the joint inflammation index,PGE2 and CRP levels (P<0.05,P<0.01).RF levels were significantly decreased in each dose of groups (P<0.01).The pathological section results of the ankle joint showed that the pathological changes in the high-dose and medium-dose IPE groups were both alleviated compared with the model group.In conclusion,I.pes-caprae extracts has significant therapeutic effect on CIA rats,which can relieve joint swelling and synovial inflammation symptoms,which may be related to the reduction of PGE2,RF and CRP expression. |
