利用3个随机引物(S453,S454,S463)对无裂栉江珧(Atrina pectinata Linaeus)的4种类型:青口、黄口、沙螺、棘螺基因组DNA进行RAPD分析,并对RAPD的实验条件行了优化研究。10~20ng的高纯度DNA用作模板,退火温度为39℃时,RAPD扩增的效率较高,扩增的带纹清晰可辨。3个引物中,引物S453和S463获得了清晰的RAPD扩增结果,引物S454未获得有效扩增,共扩增出8条清晰的DNA片段。8条清晰的类群特异或类群共享的DNA片段,能将沙螺、棘螺、油螺(青口与黄口的合称)区分开来,尚未发现能将青口与黄口分开的RAPD扩增带。 |

By RAPD analysis of four groups in Atrina pectinata Linaeus as green pen shell、yellow pen shell、thorny pen shell and scabrous pen shell,effective amplification was observed in the action of S453 or S463. As a result,some polymorphic DNA fragments of individual were revealed,8 clear group specific or group shared DNA fragments were selected for characteristic bands to preliminarily distinguish the three groups thorny pen shell,scabrous pen shell and oilily pen shell (a general designation of green pen shell and yellow pen shell).No characteristic band can be used to distinguish green pen shell from yellow pen shell. |
