用6-二甲基氨基嘌呤(6-DMAP)抑制马氏珠母贝(Pinctada martensii Dunker)受精卵的极体释放获得三倍体。在26℃条件下,进行了不同浓度梯度的6-DMAP(40mg/L、60mg/L、80mg/L、100mg/L、120mg/L)和不同的处理持续时间(5min、10min、15min、20min、25min)的诱导实验,抑制第一和第二极体的三倍体诱导率最高分别达32.4%和57.7%。随着处理强度的加大,三倍体诱导率增高,但孵化率降低。结合三倍体诱导率和幼虫孵化率,以6-DMAP浓度80mg/L、处理持续时间10min最好。 |

6-Dimethlaminopurine (6-DMAP) was used for inhibiting polar body of the zygotes to induce triploid pearl oyster (Pinctada martensii D.).At 26℃,triploid induction trails were carried out using 6-DMAP treatment at different concentration (40 mg/L、60 mg/L、80 mg/L、100 mg/L、120 mg/L) and different treatment duration (5 min、10 min、15 min、20 min、25 min).The highest induction rate of 1 pb-triploid and 2pb-triploid were 32.4% and 57.7%. Along with the increase of the treatment intensity the induction rate of triploid increased and the hatching rate reduced. Combining to the induction rate of triploid and hatching rate, the best treatment concentration of 6-DMAP was 80 mg/L and the best treatment duration was 10 min. |
